Perforin targets the membrane surface whereupon 18 to 25 monomers self-assemble into large pores (~ 120 Å inner diameter) in a calcium dependent fashion [34,94]. There, the perforin pores allow the delivery of a cocktail of pro-apoptotic proteases, called granzymes, into the target cell cytosol.
