The ATP levels were analyzed using an in-line solid-phase extraction column coupled to a reverse-phase LC-18 column, as previously described (Jones, 1981). However, quantification of free NADH levels in the cytosol or nucleus using endogenous NAD(P)H fluorescence methods remained very challenging, not only because of the very low NADH concentration in the cytosol or nucleus, but also because the major cytosolic reduced pyridine dinucleotide is NADPH [46,47]. Genetically encoded NADH sensors are not only much brighter than endogenous NADH fluorescence, but also targetable to different subcellular compartments, therefore they are more suitable for dynamic tracing of NADH, particularly for subcellular compartments like cytosol and nucleus.
