Chitosan–chondroitin sulfate and chitosan–hyaluronate polyelectrolyte complexes: biological properties
Abstract
In this work, we compare some biological properties of a highly deacetylated chitosan to those obtained with the materials made from its polyelectrolyte complexes with various GAG’s such as chondroitin-sulfates and hyaluronic acid. The hydrolysis of the complexes by means of the specific hydrolytic enzymes is studied. Cell-adhesion and cell-proliferation on these materials is compared to that obtained with a pure chitosan material. Finally, a series of in vivo experiments is performed to test the wound-healing properties of this kind of complexes. All the results agree to show that chitosan has a protective effect against GAG’s hydrolysis by their specific enzymes but only at pH’s different from the optimal pH of the enzyme considered. In addition, they also agree to confirm that a pure chitosan material gives the best results in connection with cell-attachment and cell-proliferation or wound healing. Nevertheless, whatever the case, no adverse effect was observed with the polyelectrolyte complexes GAG’s-chitosan.

Complexation between chitosan and carboxymethyl cellulose in weakly acidic, neutral, and weakly alcaline media
2023, International Journal of Biological MacromoleculesThe interaction between carboxymethyl cellulose and partially reacetylated chitosan soluble in acidic and alkaline aqueous media is studied by light scattering and isothermal titration calorimetry in a wide pH range. It is shown that the formation of polyelectrolyte complexes (PEC) can occur in the pH range of 6–8, while this pair of polyelectrolytes loses the ability to complexation upon transition to a more alkaline medium. The revealed dependence of the observed enthalpy of interaction on the ionization enthalpy of the buffer indicates the participation of proton transfer from the buffer substance to chitosan and its additional ionization in the binding process. This phenomenon is first observed in a mixture of a weak polybase chitosan and a weak polyacid. The possibility to obtain soluble nonstoichiometric PEC by a direct mixing of the components in a weakly alkaline medium is shown. The resulting PECs are polymolecular particles in shape close to homogeneous spheres with a radius of about 100 nm. The obtained results are promising for creating of biocompatible and biodegradable drug delivery systems.
Biological MWCNT/chitosan composite coating with outstanding anti-corrosion property for implants
2023, Colloids and Surfaces B: BiointerfacesBiocompatible coatings that can protect metal implants have great potential in tissue engineering. In this work, MWCNT/chitosan composite coatings with hydrophobic-hydrophilic asymmetric wettability were facilely prepared by one-step in situ electrodeposition. The resultant composite coating exhibits excellent thermal stability and mechanical strength (0.76 MPa), benefiting from the compact internal structure. The thickness of the coating can be controlled precisely by the amounts of transferred charges. The MWCNT/chitosan composite coating demonstrates a lower corrosion rate due to its hydrophobicity and compact internal structure. Compared with exposed 316 L stainless steel, its corrosion rate is reduced by two orders of magnitude from 3.004 × 10−1 mm/yr to 5.361 × 10−3 mm/yr. The content of iron released from 316 L stainless steel into the simulated body fluid drops to 0.1 mg/L under the protection of the composite coating. In addition, the composite coating enables efficient calcium enrichment from simulated body fluids and promotes the formation of bioapatite layers on the coating surface. This study contributes to furthering the practical application of chitosan-based coatings in implant anticorrosion.
Homogeneous preparation of water-soluble products from chitin under alkaline conditions and their cell proliferation in vitro
2023, International Journal of Biological MacromoleculesThe purpose of this study was to prepare water-soluble products by homogeneous depolymerization of chitin with H2O2 under alkaline conditions and investigate their potential application in wound healing. For the first time, water-soluble products were successfully prepared using a chitin-NaOH/urea solution; the products were chitosans with molecular weights (Mw) of 3.48–33.5 kDa and degrees of deacetylation (DD) > 0.5. Their Mw, DD and yield were affected by the reaction temperature, reaction time, concentration of H2O2 and chitin DD. The deacetylation and depolymerization of chitin were achieved simultaneously. The depolymerization of chitin was caused by hydrogen abstraction of HO, whereas the deacetylation resulted from the cleavage of amide bonds by HO− and HO2−, although the latter played a more important role. All water-soluble chitosans markedly promoted the proliferation of human skin fibroblast (HSF) cells, but they inhibited the proliferation of human keratinocyte cells. For the proliferation of HSF, a low concentration of chitosans was important. In addition, water-soluble chitosans with an Mw of 3.48–16.4 kDa markedly stimulated the expression of growth factors such as PDGF and TGF-β by macrophages. Water-soluble chitosans could be used as a potential active component in wound dressings.
Surface modification of silica nonwoven fabrics for osteogenesis of bone marrow-derived mesenchymal stem cells
2022, Journal of Bioscience and BioengineeringSilica nonwoven fabrics (SNFs) with high mechanical strength and porosity are known to exhibit high cell proliferation and osteogenic differentiation potential of mesenchymal stem cells (MSCs) by morphologically mimicking the extracellular matrix (ECM). To further improve the osteoinductive ability of SNFs, it could be effective to increase the interaction between MSCs and ECM components because exogenous ECM components seem to modulate the fate of MSCs differentiation. In this study, we developed immobilization methods for ECM components, such as collagen, fibronectin, and chondroitin sulphate C on SNFs, to improve cell–matrix interactions and examined their suitability for bone tissue regeneration. Collagen and fibronectin were immobilized via physical adsorption and chondroitin sulphate C was also immobilized by the layer-by-layer method combined with chitosan on SNF surfaces to maintain the high porosity of SNFs. The treated SNFs were characterized using scanning electron microscopy, Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy. In osteogenic differentiation culture, modified SNFs showed significantly increased expression of osteogenic differentiation marker genes compared to unmodified SNFs. These results suggest that the present methods improve cell–matrix interactions and enhance the cellular functions of MSCs. We are convinced that these simple modification techniques for ECM components are effective in functionalizing various 3D fabric scaffolds possessing hydrophilic groups.
One-step electrodeposition of Janus chitosan coating for metallic implants with anti-corrosion properties
2022, Colloids and Surfaces A: Physicochemical and Engineering AspectsHydrophilic-hydrophobic Janus chitosan coating was facilely fabricated by one-step electrodeposition in HCl/ethanol solvent. The face of chitosan coating attached to the electrode demonstrates a compact structure while the other face away from the electrode demonstrates a porous structure, which endows the chitosan coating asymmetric wettability. The concentration of ethanol has a profound effect on the structure and wettability of the chitosan coating and pioneer work on the corrosion resistance of the chitosan coating on metallic implants was carried out. The growth kinetics of the Janus chitosan coating reveals that the coating grows faster and contains more pores in the presence of ethanol. The anticorrosion properties of the Janus chitosan coating were investigated on common metal implants titanium and 316 L stainless steel. The results show that the Janus chitosan coating obtained with 10 wt% ethanol electrolyte has the best anti-corrosion ability due to its compactness and hydrophobic surface. The corrosion rates of the Janus chitosan coating were reduced to 3.94 × 10−4 mm/yr and 1.77 × 10−2 mm/yr, respectively, significantly improved as comparison to the bare metal corrosion rates (1.46 × 10−3 mm/yr for bare titanium and 3.00 × 10−1 mm/yr for bare stainless steel). The present work provides a simple way to prepare Janus chitosan coating and demonstrates its great potential in implant anticorrosion.
Application of chitosan-based nanoparticles in skin wound healing
2022, Asian Journal of Pharmaceutical SciencesThe rising prevalence of impaired wound healing and the consequential healthcare burdens have gained increased attention over recent years. This has prompted research into the development of novel wound dressings with augmented wound healing functions. Nanoparticle (NP)-based delivery systems have become attractive candidates in constructing such wound dressings due to their various favourable attributes. The non-toxicity, biocompatibility and bioactivity of chitosan (CS)-based NPs make them ideal candidates for wound applications. This review focusses on the application of CS-based NP systems for use in wound treatment. An overview of the wound healing process was presented, followed by discussion on the properties and suitability of CS and its NPs in wound healing. The wound healing mechanisms exerted by CS-based NPs were then critically analysed and discussed in sections, namely haemostasis, infection prevention, inflammatory response, oxidative stress, angiogenesis, collagen deposition, and wound closure time. The results of the studies were thoroughly reviewed, and contradicting findings were identified and discussed. Based on the literature, the gap in research and future prospects in this research area were identified and highlighted. Current evidence shows that CS-based NPs possess superior wound healing effects either used on their own, or as drug delivery vehicles to encapsulate wound healing agents. It is concluded that great opportunities and potentials exist surrounding the use of CSNPs in wound healing.
