The preparation of functionalized amines and amides using benzotriazole derivatives and organozinc reagents

Abstract

Secondary and tertiary amines 6 are conveniently prepared in good yields by reacting organozinc halides with adducts 4, which are derived from the corresponding amines, aldehydes and benzotriazole. The use of organozinc reagents enables the introduction of nitro, cyano or ester functionalities into amine mainframes. This methodology was also used to prepare functionalized amides 8.

The reactions of organozinc halides with benzotriazole derivatives 1 enable the introduction of nitro, cyano or ester functionalities into secondary and tertiary amine or amide mainframes 2.

      In our preceding study,12b we mentioned that commercial tetrahydrofuran solutions of aromatic organozinc reagents18 are inefficient in the three-component coupling. In order to verify that arylzinc reagents are particularly ineffective when used as tetrahydrofuran solutions, we prepared 4-methoxyphenylzinc bromide in tetrahydrofuran, starting from 4-bromoanisole, by a metal–halogen exchange using butyllithium and a transmetalation by means of zinc chloride.19 This organozinc reagent did not lead to satisfactory results, only traces of its coupling product with benzaldehyde and piperidine being detected in the reaction mixture, even after several hours stirring at room temperature.
