The first attempt to detect DNA repair in mitochondria analyzed removal of UV-induced damage, the prototypical Nucleotide Excision Repair (NER) substrate, from mtDNA and found that these lesions were not removed at significant rates in mammalian [115] or yeast cells [116], leading to the long-standing notion that mitochondria lacked DNA repair systems. In fact, to date no clear evidence of canonical NER in mitochondria has been published (for mechanistic details on nuclear NER, please see chapter “Damage removal and gap filling in nucleotide excision repair” by Kemp in this volume).
N-methylpurines are heterogeneously repaired in human mitochondria but not evidently repaired in yeast mitochondria

Nucleotide excision repair, which plays a primary role in repairing helix-distorting bulky DNA lesions in nDNA, has been shown to be inactive in the mitochondria of both human [12] and yeast [13] cells. However, certain levels of BER, mismatch repair, homologous recombination and nonhomologous end-joining appear to be present in mitochondria (for a recent review, see Ref. [14]).
