Melittin promotes exocytosis in neuroendocrine cells through the activation of phospholipase A<inf>2</inf>
2010, Regulatory PeptidesCitation Excerpt :The intracellular effector is thus likely to be a Ca2+-regulated protein. Indeed, to access its substrate, the cytoplasmic PLA2 has to be translocated from the cytosol to membranes and this translocation is known to be Ca2+-dependent [28]. However, Ca2+ per se is only a weak activator of PLA2, as seen here by the low amount of arachidonate release induced by elevated Ca2+ concentrations in permeabilized chromaffin cells (Figs. 2–3).
Control of free arachidonic acid levels by phospholipases A<inf>2</inf> and lysophospholipid acyltransferases
2009, Biochimica et Biophysica Acta - Molecular and Cell Biology of LipidsTears and Their Secretion
2005, Advances in Organ BiologyCitation Excerpt :Molecular cloning has revealed that there are three families of PLCs: β, δ, and γ with molecular weights ranging from 85 to 154 kDa. Within each family, there are several cloned subtypes designated by adding Arabic numerals after the Greek letters (Cockroft, 1992). All three types of PLC catalyze the hydrolysis of the three common inositol‐containing phospholipids: phosphatidylinositol (PI), phosphatidyl 4‐monophosphate (PIP), and phosphatidyl 4,5‐bisphosphate (PIP2), with PIP and PIP2 the preferred substrates.
Signaling to migration in neutrophils: Importance of localized pathways
2003, International Journal of Biochemistry and Cell BiologyProtein kinase C α associates with phospholipase D<inf>1</inf> and enhances basal phospholipase D activity in a protein phosphorylation-independent manner in human melanoma cells
2003, Journal of Investigative DermatologyCitation Excerpt :The cell debris was scraped into an Eppendorf tube (1.5 mL) and kept on ice, and a solution of CHCl3/HCl (1:0.006, vol/vol) (400 μL) was added to each tube. Phase separation, lipid extraction, and thin layer chromatography analysis were carried out as described previously (Cockcroft, 1992). Radioactivity was quantitated using a Bioimaging analyzer (BAS, 2000; Fuji Film, Tokyo, Japan), and the PLD activity was expressed as the percentage of radioactivity in phosphatidylethanol relative to the total lipid radioactivity found in a given lane.
