2013, PlasmidCitation Excerpt :How were the elements that negatively influence replication initiation first identified (reviewed in Novick, 1987; Nordstrom, 1990)? In an approach referred to as incompatibility (Inc+) testing, fragments of minimal replicons were screened to identify factors/sequences that inhibit the replication of the plasmid from which they originated; the original analysis of this type was first done for mini F plasmid (Tolun and Helinski, 1981). Using similar methodology it was determined that π/γ ori interactions are the source of incompatibility in R6K (Filutowicz et al., 1985b; McEachern, 1987).

A single iteron from a plasmid of a given incompatibility group, when cloned into a second compatible plasmid, confers on the latter incompatibility with the first plasmid that donated the iteron (Tolun and Helinski, 1981). This observation suggests that competition for binding to the same initiator protein could be a basis of incompatibility.
