Sarcophine—a new toxin from the soft coral Sarcophyton glaucum (Alcyonaria)

Abstract

The soft coral Sarcophyton glaucum is avoided by predators and has a toxic effect on fish. From the hexane extract of the dried coral a new diterpene was isolated. This compound was given the name sarcophine. Mass spectra and elemental analysis gave its formula as C30H25O3. The functional groups of sarcophine are: (a) αβ-unsaturated lactone, (b) epoxide, (c) two additional double bonds. Thiol addition to the αβ-unsaturated lactone was investigated. Sarcophine seems to be the main toxic substance of Sarcophyton glaucum. Lethality tests, using Gambusia affinis showed that the ld50 of the toxin after 3 hr is 3 mg per liter. Sarcophine was toxic to mice, rats and guinea pigs and showed strong anti-acetylcholine action on the isolated guinea pig ilcum. Sarcophine was also a competitive inhibitor of cholinesterase in vitro.

    The appearance of an oxygenated quaternary carbon signal at δC 73.2 and a methyl signal at δC 28.9, accompanying with the disappearance of the typical 13C NMR signals for the exocyclic olefin at Δ8,19 in 2 clearly indicated the hydroxylation occurred at C-8.     These animals live exposed to predators in their environment, thus they rely mainly on their essential toxic secondary metabolites to repel potential attackers (Bilasy et al., 2006). Ever since its discovery, SN has undergone extensive investigations for promising biological activities, especially for its antitumour potential (Ne’eman et al., 1974; Liang and Guo, 2013; Zubair et al., 2016; Chao et al., 2017). However, the toxicity of SN remains an obstacle against introducing it for human clinical trials (Bernstein et al., 1974).
