Characterization of subtilisins by isoelectric focusing and polyacrylamide gel electrophoresis

Abstract

  • 1.

    1. In a search for additional criteria for the differentiation of subtilisins, the applicability of isoelectric focusing and polyacrylamide gel electrophoresis was tested and the isoelectric points of subtilisin Carlsberg and Novo were re-examined.

    2. The determination of the electrophoretic characteristics of subtilisins is hindered by their high rate of autolysis and by the formation of enzyme-breakdown product complexes.

  • 3.

    3. However, subtilisin Carlsberg and Novo could be differentiated by disc gel electrophoresis using a pH 4.3 system and by isoelectric focusing, if the native and inhibited enzyme analysis were combined. Isoelectric points were 6.7 for subtilisin Carlsberg and 8.1 for subtilisin Novo.
