Developments in the use of chromatographic techniques in marine laboratories for the determination of halogenated contaminants and polycyclic aromatic hydrocarbons

Chromatography has been an important tool in marine laboratories. Since the 1960s, marine laboratories have been involved in the analysis of polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs), polycyclic aromatic hydrocarbons (PAHs), and brominated flame retardants (BFRs). Column chromatography and liquid chromatography (LC) techniques have been used, mainly in the clean-up phase, while gas chromatography (GC) has been used extensively in the final determination of these contaminants. Developments have been observed from the use of packed GC columns, via capillary columns to the use of heart-cut multi-dimensional GC and comprehensive multi-dimensional GC. The progress made in interlaboratory studies and the availability of certified reference materials are discussed.

Methodology of PAHs analysis was strongly affected by levels of development of chromatographic methods. At the beginning, a separation of BaP isomers by PC and CC was practically impossible [24]. With regard to complex mixtures of PAHs, the presence of a variety of interfering substances and the need to assess correctly real concentrations of the most dangerous compounds at minimum, it was necessary to overcome problems regarding the resolution of so-called ‘benzpyrene fraction’ which consisted at this time of BaP and its isomers BeP, BkF, BbF and Per.
Formation, factors affecting concentrations, legal limits and occurrence of polycyclic aromatic hydrocarbons in smoked meat products and smoke flavour additives are briefly reviewed. The most used techniques such as thin-layer chromatography (TLC), gas chromatography (GC) and high-performance liquid chromatography (HPLC) are evaluated. Also, sample preparation, pre-separation procedures, separation and detection systems being used for determination are discussed with emphasis to latest development in applied food analysis and the chosen data regarding the concentration of polycyclic aromatic hydrocarbons in smoked meat products and smoke flavour additives are summarised.
Comparison of methods for separating polycyclic aromatic hydrocarbons by high-performance thin-layer chromatography

A comparison is made of different stationary phases (silica gel, cellulose, acetylated cellulose, and 3-aminopropyl-, ethyl-, octyl-, octadecyl-, and diphenylsilanized silica gel), mobile phases, and development techniques (conventional, continuous, and multiple) for the separation of environmentally important polycyclic aromatic hydrocarbons by high-performance thin-layer chromatography. Reversed-phase separations using octadecylsilanized silica gel and methanol—water or methanol—water-based ternary solvent systems were found to give the best separations. A new development technique, multiple development with application of the mobile phase further up the plate at each successive development, provided superior resolution of the polycyclic aromatic hydrocarbons compared with conventional multiple development.
Qualitative identification of polycyclic aromatic hydrocarbons by high-performance thin-layer chromatography and fluorescence scanning densitometry

A procedure is described for the accurate measurement of emission response ratios for polycyclic aromtic hydrocarbons using fluorescence scanning densitometry and reversed-phase high-performance thin-layer chromatography. By optimizing instrumental conditions and using perylene as a reference standard, the emission response ratios can be reproduced to better than 3% relative standard deviation on a day-to-day basis. The emission response ratios enable the optimum detection conditions for sensitivity or selectivity to be easily identified. The normalized emission response ratios can be used to improve the confidence in the identification of a particular compound made by coincidence of retention in the chromatographic system. The method has been applied to the identification of polycyclic aromatic hydrocarbons in samples of laboratory dust and a diesel engine exhaust particulate sample.
Determination of polycyclic aromatic hydrocarbons in automobile exhaust by means of high-performance liquid chromatography with fluorescence detection

A chromatographic method has been developed and applied to the determination of polycyclic aromatic hydrocarbons (PAHs) in particulate matter in automobile exhaust, in petrols, and in crankcase oils. The PAHs were purified from other organic compounds by thin-layer chromatography, separated by high-performance liquid chromatography, and measured by means of on-line fluorescence detection. The identities of the PAHs were verified by comparing the emission spectra obtained by a stop-flow technique with those of standard PAHs.
Determination of benzo[a]pyrene in smoked, cooked and toasted food products

Samples of smoked, cooked and toasted food products, commercially available in Italy, and some samples of oils and fats were analysed for benzo[a]pyrene. In the commercial samples, benzo[a]pyrene was detected in approximately 73% of the samples, at levels ranging from 0–01 to 9·51 μg/kg. In electrically-broiled foods, the benzo[a]pyrene content ranged from 0 to 0·05 μg/kg. Levels of benzo[a]pyrene found in the food products analyzed were lower than levels reported by other investigators for comparable food commodities.
