Inhibition of secreted LPL activity by TNFα has been observed in mouse peritoneal macrophages but not in J774.1, BMDM and human monocyte-derived macrophages [252,253,255]. Interferon γ decreases macrophage LPL at least in part via a reduction in LPL synthesis and gene transcription, possibly via CK2 and PI3K signaling and regulation of Sp1/Sp3 binding [256,257].

The 2×106 cells/well were seeded in minimal essential medium (MEM) containing 0.1% fatty acid-free bovine serum albumin and incubated with [3H]CE-Intralipid for 24 h. [3H]cholesteryl oleate ([3H]CE) was evaporated and 1 ml Intralipid in PBS (1 mg triglyceride/ml) was added and cosonicated for 6×30″, using a 4-mm tip and the Braun-Sonic 3000 sonicator (Braun, Melsungen, Germany), at 50% output [16].
