Interference of parathion with mitochondrial bioenergetics

Abstract

The organophosphorus insecticide parathion depresses the phosphorylation efficiency of mitochondria as inferred from the decrease of RCR and ADP / O ratios. The transmembrane potential (Δψ) developed by energized mitochondria, and depolarization upon ADP addition are also decreased. Furthermore, repolarization is delayed and resumes at a slower rate. The inhibitory action of parathion on phosphorylation efficiency could be related with the following findings: (1) a direct effect on the succinate dehydrogenase-ubiquinone segment of the redox chain; (2) a direct action on the ATP synthetase complex; (3) partial inhibition of the phosphate transporter.

      The increase of oxygen after addition of ADP referred to state III respiration and state IV occurs after phosphorylation of all the ADP to ATP in slower rate. The phosphorylation efficiency of mitochondria inferred from ratios of state III to state IV respiration (RCR) and ATP to oxygen consumption (ADP/O) was decreased by parathion (Moreno and Madeira, 1990). In another study, respiration rate and RCR ratio were not altered significantly in rats exposed to monocrotophos (Venkatesh et al., 2009).

      Retardation of photosynthesis, accelerated respiration and altered pigment fluorescence and Photosystem II (PS II) activity of various algae and cyanobacteria have been reported under various insecticide stress (Mohapatra and Schiewer, 2000; Yao et al., 2006; Pan et al., 2008; Li et al., 2010; Chen et al., 2011). There are also reports that the photosynthetic electron transport in thylakoid membrane as well as the electron transport in mitochondrial membrane are affected differently by OP insecticide toxicity (Moreno and Madeira, 1990; Mohapatra et al., 1996; Panda et al., 1998; Li et al., 2010).       A clear inhibitory effect on succinate dehydrogenase is induced by both the insecticides in vitro, suggesting the inhibition of electron flow through the electron transport chain, thus the insecticide-induced depression of phosphorylation efficiency of isolated mitochondria is mainly due to the inhibition of oxygen consumption and inhibition at complex II. Similar observations were reported for the insecticide-induced mitochondrial dysfunction in vitro in rats [19]. Under in vivo conditions, state III respiration and state IV respiration decreased, affecting RCI, suggesting absence of uncoupling mechanism of action in these insecticides.
