Catalytic physiological amyloids

2022, Trends in Chemistryβ-Amyloid fibrils catalyze neurotransmitter degradation
2021, Chem CatalysisCitation Excerpt :Specifically, Figure 4 shows that Aβ42 fibrils catalyze acetylcholine hydrolysis, as well as oxidation of dopamine and adrenaline, two prominent neurotransmitters. Figure 4A depicts the effect of Aβ42 fibrils and monomers upon hydrolysis of acetylthiocholine (AtCh), a well-known acetylcholine surrogate.48 Disruption of acetylcholine-mediated signaling is one of the hallmarks of AD, linked to memory loss and known as the cholinergic hypothesis.49
Laboratory analysis of chemical warfare agents, adducts, and metabolites in biomedical samples
2020, Handbook of Toxicology of Chemical Warfare AgentsFacile synthesis of 1,2-dione-containing abietane analogues for the generation of human carboxylesterase inhibitors
2018, European Journal of Medicinal ChemistryCitation Excerpt :Once established, data were plotted using Prism (GraphPad, La Jolla, CA) and Ki values were determined from derived curve fits. A determination of the inhibition of hAChE or hBChE was undertaken in a similar fashion except that acetylthiocholine or butyrylthiocholine were used as a substrates, respectively [32,33]. Inhibitors were assayed at a single concentration (5 μM).
Radiolabelled soman binding to sera from Rats, Guinea Pigs and Monkeys
2018, Toxicology LettersCitation Excerpt :NMR analysis of the toluene extract containing 3H-soman indicated that the soman in this extract was 84.7% pure as compared to NMR analysis of the non-labeled samples of soman. The specific activity (114 mCi/mmole) of the 3H-soman in the toluene extract was determined by scintillation counting of the disintegrations per min (dpm: 1 mCi = 2.22 × 109 dpm) of an aqueous solution of the toluene extract and quantitative bioasssay of the amount of soman in this extract from its inhibition of acetylcholinesterase in comparison to a non-radioactive soman standard using a microtiter-plate modification (Doctor et al., 1987) of the Ellman assay. Blood was collected from rats and guinea pigs by cardiac puncture immediately after euthanasia of these animals.
