Microtiter assay for acetylcholinesterase

Abstract

A microtiter plate adaptation of the classical Ellman colorimetric procedure for measurement of acetylcholinesterase activity is described. This method permits use of an enzyme-linked immunosorbent assay plate reader for rapid analysis of multiple samples and is particularly suitable for analysis of acetylcholinesterase activity on sucrose gradients. The novel procedure is rapid and sensitive and does not require use of radioactive material.

    NMR analysis of the toluene extract containing 3H-soman indicated that the soman in this extract was 84.7% pure as compared to NMR analysis of the non-labeled samples of soman. The specific activity (114 mCi/mmole) of the 3H-soman in the toluene extract was determined by scintillation counting of the disintegrations per min (dpm: 1 mCi = 2.22 × 109 dpm) of an aqueous solution of the toluene extract and quantitative bioasssay of the amount of soman in this extract from its inhibition of acetylcholinesterase in comparison to a non-radioactive soman standard using a microtiter-plate modification (Doctor et al., 1987) of the Ellman assay. Blood was collected from rats and guinea pigs by cardiac puncture immediately after euthanasia of these animals.
