Lageveränderungen der Sauerstoffbindungskurve in Abhängigkeit von den intraerythrocytären Kationen- und Hämoglobinkonzentrationen

Zusammenfassung

Zur Untersuchung des Einflusses der intracrythrocytären Kationen- und Hämoglobinkonzentrationen auf das Sauerstoffbindungsverhalten wurden jeweils 1 ml Blut mit 2 ml einer hypo-, iso- oder hypertonen Ringer- bzw. Saccharoselösung vermischt. Von solchen Blut-Lösungs-Gemischen wurden folgende Parameter bestimmt: die Erythrocytenzahl, der sog. mechanische und chemische Hämatokrit, das Intercellularvolumen und der Überstand abzentrifugierter Erythrocyten, das mittlere Erythrocytenvolumen und der mittlere Erythrocytendurchmesser, die mittlere intraerythrocytäre Hämoglobinkonzentration, die intra- und extraerythrocytäre Naund K-Konzentration, der pH-Wert und als Maß für die Lage der Sauerstoffbindungskurve der Halbsättigungsdruck.

Die ermittelten Halbsättigungsdrucke lassen weder eine direkte Abhängigkeit von den intracellulären Na- oder K-Konzentrationen noch von den Gesamtkationenkonzentrationen erkennen. Ein einfacher Zusammenhang zwischen den Halbsättigungsdrucken und den intracellulären Hämoglobinkonzentrationen war ebenfalls nicht festzustellen.

Demgegenüber besteht eine lineare Beziehung zwischen den Halbsättigungsdrucken und den Quotienten aus intracellulärer Gesamtkationenkonzentration und Hämoglobinkonzentration in der Weise, daß der Halbsättigungsdruck mit Zunahme des Quotienten linear größer wird.

Die Beeinflussung des Sauerstoffbindungsverhaltens durch die Anzahl der Kationen, die pro Hämoglobinmolekül vorhanden sind, hat vermutlich eine grundsätzliche Bedeutung. Mit Hilfe des Quotienten lassen sich nach Literaturangaben bisher in ihrer Genese ungeklärte Variationen der Sauerstoffbindungskurven von Tierblut, Anämienblut und Fetalblut deuten.

Summary

In order to investigate the effect of intraerythrocytic eation and haemoglobin concentrations on oxygen dissociation, 2 ml amounts of hypotonic, isotonic and hypertonic Ringer's and saccharose solutions were set up, and 1 ml of blood was added to each. The following parameters were determined on the mixtures: erythrocyte count, the so-called mechanical and chemical haematocrits, the intercellular volume and the supernatant from the centrifuged erythrocytes, mean cell volume, mean cell diameter, mean cell haemoglobin concentration, intra-erythrocytic and extra-erythrocytic sodium and potassium concentrations, pH value, and, as a characteristic for the position of the oxygen-dissociation curve, the half-saturation pressure.

The half-saturation pressures measured failed to reveal a direct relationship with the intracellular sodium or potassium concentrations or with the total cation concentration. Nor was it possible to establish a simple relationship between the half-saturation pressures and the intracellular haemoglobin concentrations.

There is, however, a linear correlation between the half-saturation pressures and the quotients of the total intracellular cation concentration and haemoglobin concentration in such a manner that a rise in the quotient is accompanied by an increase in the half-saturation pressure.

Presumably the effect of the number of cations present per molecule of haemoglobin is of fundamental significance in oxygen-dissociation. This quotient can now throw some light on the hitherto unexplained cases of variation in the oxygen-dissociation curve in animal, anaemic and foetal blood described by various authors.
