The effects of local treatment with 1% and 3% vitamin A acid (VAa) on Guinea pig epidermis are investigated by electron microscopy.

The first noticeable changes consist in a prominent appearance of the nucleoli in all epidermal layers.

After 3 days of daily treatment with VAa the number of tonofilaments, desmosomes, keratinosomes and keratinohyalin granules is greatly reduced. At this time ribosomes, mitochondria, rough surfaced endoplasmic reticulum and Golgi complexes indicate strong stimulation of metabolic (secretory) activity throughout the whole epidermis. Vacuoles containing homogeneous, electron-dense material appear in the cytoplasm of the cells and in the intercellular space. These changes are more pronounced after treatment with 3% than with 1% VAa.

Local treatment with VAa for 8 days shows the reappearance of numerous keratinosomes and large amounts of keratohyalin.

The major effect of VAa on the epidermis consists in the inhibition of normal keratinization and in the induction of an atypical differentiation while the proliferative activity is increased.

Ultrastrukturelle Veränderungen der Meerschweinchenepidermis unter lokaler Einwirkung von 1% iger bzw. 3% iger Vitamin A-Säure (VAS) werden beschrieben.

Als erste Veränderung wird bereits 6 Std nach einmaliger Applikation der VAS ein auffälliges Hervortreten der Nucleolen beobachtet.

Nach 3 tägiger Behandlung mit VAS ist der Bestand der Epidermis an Tonofilamenten, Desmosomen, Keratinosomen und Keratohyalin gegenüber der Norm stark vermindert. Dabei finden sich im Cytoplasma der Epidermiszellen aller Schichten die Zeichen einer stark erhöhten Syntheseleistung: Ribosomen, Mitochondrien, granuläres endoplasmatisches Reticulum, Golgi-Komplexe. Gleichzeitig treten intracellulär Vacuolen mit homogenem, elektronendichtem Material auf; morphologisch gleichartiges Material enthält der verbreiterte Intercellularraum. Die genannten Veränderungen sind bei Verwendung 3% iger VAS in ausgeprägterem Maße nachweisbar als bei 1% iger VAS.

Nach 8 tägiger Behandlung treten als Charakteristica einer regelrechten Verhornung wieder zahlreiche Keratinosomen und grobtropfiges Keratohyalin in Erscheinung.

Als wesentliche Wirkung der VAS auf die Epidermis wird neben der Stimulation der Proliferation die Hemmung der normalen Ausdifferenzierung und Verhornung angesehen, bei gleichzeitiger Umdifferenzierung der Epidermis.
