Summary
We have established the HindIII physical map of a cloned 290 kilobase fragment of the Rhizobium meliloti 2011 pSym megaplasmid. The cloned fragment, which contains nodulation genes as well as the nitrogenase structural genes (nifHDK), has been shown to be colinear with the corresponding genomic region. Using transposon mutagenesis we have demonstrated that a region which is located more than 200 kb from the nifHDK operon on pSym is essential for symbiotic nitrogen fixation.
