Zusammenfassung
Die Verwendung von (C14)-markiertem Serotonin ermöglicht es, die Freisetzung dieser Substanz aus den Thrombocyten im Plasma zu messen, wenn eine Antigen-Antikörperreaktion im Suspensionsmedium abläuft. Dieses System wurde verwendet, um die Freisetzung von Serotonin nach Zusatz zweier möglicher Antigene — encephalitogener Faktor (EF) und Eialbumin — zum Plasma zu messen. Auf diese Weise läßt sich das Vorhandensein von Antikörpern, die anderen Nachweismethoden entgehen, demonstrieren. Zugabe von EF und Eialbumin zu MS-Plasma setzt im Gegensatz zu den Kontrollen Serotonin frei; EF ist in Fällen von MS aktiver als Eialbumin. Eine Gruppe anderer neurologischer Erkrankungen zeigte jedoch das gleiche Verhalten. Die Bedeutung der in Frage kommenden Antikörper wird diskutiert.
Summary
Using C14 labelled serotonin it is possible to measure the release of this substance from blood platelets in plasma when an antigen-antibody reaction takes place in the medium in which they are suspended. The system has been used to measure the release of serotonin when two possible antigens — encephalitogenic factor (EF) and ovalbumin — are added to plasma. The method is one which may reveal the presence of an antibody not demonstrable by other means. EF and ovalbumin both cause release of serotonin when added to MS plasma as compared with controls; and EF is more potent than ovalbumin in cases of MS. However, a group of non-MS neurological cases also showed release. The significance of the antibodies concerned is briefly discussed.
