Abstract
Here we present a microbiology microfluidic device that permits the growth of a bacterium on-chip. Our device was fabricated from Polydimethylsiloxane using an SU-8 photoresist positive master. The device consists of a microfabricated capillary whereby a suspension of bacterial cells are flowed through and incubated. Incubation of an Escherischia coli expressing a green fluorescent protein was successfully monitored optically and by fluorescence measurements over a 4–5 hour period. The system confers several advantages in comparison to conventional bulk growth techniques such as real-time monitoring, reduction in inoculum volume and a considerable reduction of biological waste.
