Enzyme Inhibition Sensor for Organophosphorus Pesticides Analysis

Uses of Immobilized Biological Compounds

Part of the book series: NATO ASI Series ((NSSE,volume 252))

Abstract

The study of the inhibiting action exerted by organophosphorus pesticides on the esterase enzymes is of great interest, due to the potential dangerous activity of pesticides and to their largescale use in agriculture. Different biosensors for the determination of pesticides were developed by several researchers in the recent years, basing on this behaviour. The biosensor developed by us consists of a Clark oxygen electrode, modified with a dialysis membrane, placed over it, containing immobilised two biological mediators: butyrylcholinesterase and choline oxidase. A comparison with the results obtained by a similar previously published procedure, characterised by butyryl- (or acetyl-) Cholinesterase free in solution, is also reported. A new enzyme sensor to determine organophosphorus pesticides is also described, constituted by a hydrogen peroxide amperometric electrode modified by a superimposed nylon membrane containing, chemically immobilised, two biological mediators (butyrylcholinesterase and choline oxidase). Some applications to authentic matrices of environmental or of agricultural interest are reported and discussed.
