Zusammenfassung

In einer Übersicht wird zur Pharmakokinetik, zu pharmakologischen Effekten und zur Toxikologie der topischen Benzoylperoxid-Behandlung Stellung genommen. Benzoylperoxid ist antimikrobiell wirksam und weist eine hemmende Wirkung auf die Zellproliferation in der Talgdrüse auf. In der Haut wird es schnell zu Benzoesäure abgebaut. Die akute und chronische Toxizität sind bemerkenswert gering. Eine mutagene oder embryotoxische Wirkung wurde nicht nachgewiesen. Unter bestimmten Versuchsbedingungen kann Benzoylperoxid tumorpromovierend wirksam werden. Dies gilt jedoch nicht im Zusammenhang mit dem an der Haut bedeutsamsten Kanzerogen Licht. Klinische Hinweise auf Tumorbildungen unter Benzoylperoxid-Behandlung finden sich in der Literatur nicht. Gering ist die sensibilisierende Wirkung bei der Aknebehandlung, sehr hoch ist sie indessen bei der Ulcus cruris-Behandlung. Die klinische Indikation ist im wesentlichen die Acne vulgaris.

Summary

In a survey, the pharmacokinetics, pharmacological effects and toxicology of topical treatment with benzoyl peroxide are commented. Benzoyl peroxide acts antimicrobially and possesses inhibiting effects on cell proliferation within the sebaceous gland. In the skin it is rapidly decomposed to benzoic acid. Toxicity, both acute and chronic, is remarkably low. Mutagenic or embryotoxic effects were not demonstrated. Under certain experimental conditions, benzoyl peroxide may act tumor-promoting. This, however, is not valid in connection with the most important skin carcinogenic, i.e. light. Clinical indications of tumor formation during treatment with benzoyl peroxide are not found in literature. The sensitizing effect is minimal in acne therapy, yet very great in the therapy of leg ulcers. The chief indication is common acne.
