Abstract
We assessed the inhibition of tumour cell attachment by local treatment with phospholipids to the peritoneum in naked mice and the influence of phospholipids on adhesion of gastric cancer cells (NUGC-4) on extracellularmatrix components, in vitro. In 90 female BALB/C nu/nu mice gastric cancer cells (1 × 106) were applicated intraperitonealy suspended in either normal saline (controls), phospholipid 1.5% (PL75) or 3% (PL150). After 30 days, the extent of peritoneal carcinosis (mm2, ml) and the Peritoneal Cancer Index (PCI) were evaluated. The intraperitoneal tumour volume could be reduced by phospholipid 3% up to a value of 0.48 ± 0.09 ml compared to 0.9 ± 0.1 ml in the control group (p = 0.04). The mean area of tumour adhesion amounted up to 164 ± 32.8 mm2 after treatment with phospholipid 3% (control: 245 mm2 ± 29.3; p = 0.049). The peritoneal cancer index was reduced from 21.7 ± 0.77 (control) up to 14.3 ± 1.07 with phospholipid 3% (p = 0.0001). We found a prolonged survival rate after treatment with phospholipid 3%. Microtiter plates were coated with collagen IV, laminin and fibronectin for in vitro adhesion experiments. Cell adhesion to laminin, collagen IV and fibronectin could be reduced significantly. Phospholipids may be an efficacious tool to reduce peritoneal tumour cell adhesion after resection of gastric cancer.
