Abstract

Neoplastic transformation is a multistep process involving the arrangement, amplification, mutation or deletion of specific genes. Several mechanisms may account for these gene modifications; they include point mutations and chromosome mutations (see Barrett, this volume). Genotoxic agents may be responsible for primary or secondary events which play a critical role in cell transformation by the production, not only of point mutations, but also of heritable changes through gene modifications and rearrangements. Agents that interact with DNA structure, chromosomes or even interfere with DNA metabolism are potentially genotoxic.
