Abstract
Most of the studies described in this chapter were carried out with a series of multidrug-resistant (MDR) derivatives of the human KB carcinoma cell line. KB cells, used for selection of the MDR cell lines, represent a subclone of human HeLa cells (Nelson-Rees and Falndermeyer, 1976), as indicated by their karyotype and cell surface markers. These cells were chosen for the study of MDR because they are highly sensitive to most chemo-therapeutic drugs, clone with high efficiency (>20%), grow rapidly (t l/2 < 20 hr), and can be subcloned in 7–10 days. MDR derivatives of KB cells were obtained by selection with colchicine, vinblastine, or Adriamycin, which represent three structurally dissimilar groups of cytotoxic drugs associated with multidrug resistance.
